Doktorandplatserna avser två års utbildningsbidrag och två års doktorandanställning, alternativt fyra års doktorandanställning, och beräknas leda fram till doktorsexamen.

Eosinophilic eosophagitis - new mystical disease

Description of the research project
The focus of this project is basic immunological studies of Eosinophilic esophagitis, a newly emerged disease featuring infiltration of the esophagus by eosinophils and gastrointestinal symptoms including difficulty in swallowing.The cause of the disease is unknown but there is an association with allergy to food and inhalant allergens.Three main questions will be addressed: 1) Is eosinophilic esophagitis an infectious disease?2) Do eosinophils derived from eosinophilic esophagitis patients differ depending upon if the patient is allergic or not allergic?In other words, are there different forms of eosinophilc esophagitis?3) Which danger signals are recognized by eosinophils in eosinophilic esophagitis?A non-cultivatable infectious agent will be searched for by PCR analysis of conserved microbial DNA sequences in tissue biopsies followed by sequencing.Eosinophils will be characterized regarding the expression of > 20 molecules, cytokine profiles in blood determined, and patients characteristics assessed, and all data processed by multivariate methods of pattern recognition to establish if there are subtypes of eosinophilic esophagitis, and how their patterns compare to those of food or inhalant allergic individuals.Finally the reactivity of eosinophils to danger signals (allergens, necrotic cells, formylated peptides, parasite extracts) will be investigated, and the activation parameters evoked analyzed (chemotaxis, respiratory burst, intracellular calcium transients, generation of cytokines and eicosanoids).
