The research area and duties
The project is part of a project funded by the Swedish Research Council, aiming to clarify the role of cooperative phenomena and hierarchical ordering in striated muscle function in health and disease (e.g. hereditary cardiomyopathies). The project will be carried out in collaboration between A. Månsson and S. Tågerud at Linnaeus University, H. Linke (Lund University, Sweden), D. Rassier (McGill University, Canada) and J.A. Spudich (Stanford University, USA). The focus will be on development of a mouse myotube based expression and purification system for human striated muscle myosin II. The expressed proteins will be used in various in vitro assays including a novel small-ensemble force measurement system, for studies of hypertrophic cardiomyopathy. The work will be performed in Kalmar, but extensive interaction with labs of collaborators is expected. The postdoc, will work in tight collaboration with a PhD student with focus on biochemistry and biophysics of isolated muscle proteins and a postdoc with major focus on a novel force measurement system. Key experimental techniques, in addition to the newly developed expression system, will be fluorescence microscopy and in vitro motility assays. Supporting expertise to assist the development of the expression and purification system exist in house.

Qualification requirements
Applicants should have a PhD degree in a subject area of relevance for the project (biochemistry, molecular biology, cell biology, biomedical engineering etc.).

Other assessment grounds
Experience of experimental work using mammalian protein engineering and expression systems is a merit including skills like site directed mutagenesis, DNA cloning, large scale mammalian cell culturing, protein expression and purification. Previous studies of molecular motors and/or muscle are judged favorably. In addition, the Post Doc should have a high proficiency in written and spoken English, exhibit high problem solving skills, and have the ability to work independently as well as interact well in a research group.
